I need to perform a sterility test by filtration (USP 71) on a solution containing some particles (transport solution from fresh heart recovered for heart valves transplant). So the particles seems to be red blood cell, some proteins, lipids and all kind of stuff like that.

I want to make a transposon library in a gram negative bacteria. The genome size in ~1.9 Mb and average gene size is 1 kb. Does anyone know how I calculate the theoritical number of transposon mutants I need to knockout every gene in this bacteria?

I know someone trying to maintain a cell culture of some sensitive microbes but they are having trouble with pH control. I'm just a chemist but could use some more suggestions. Pond water, ca. pH 7-7.2 and mud is harvested. It contains a variety of life.
