It is said that the entire human genome is contained in 50K to 100K genes. Given that each gene codes for only one protein/enzyme, that seems quite impossible. What's the key to this? BTW, is there a more definite number for gene count these days?

I think it is easily to select bacteria fom a mixture (sewage, soil...) that can use a specific carbohydrate in addition to different ones by makeing the specific carbohydrate the only one in the substrate the bacteria are grown in.
